Lawler made his UFC debut against veteran Aaron Riley at UFC 37. Lawler was won by unanimous decision. Lawler fought again the following month at UFC 37.5, defeating Steve Berger by TKO. At UFC 40, Lawler met another veteran, Tiki Ghosn, and won in a highlight-reel knockout. Lawler later faced Pete Spratt at UFC 42 but lost by submission due to a knee ailment. Lawler won a majority decision over fellow boxing and wrestling expert Chris Lytle at UFC 45.

Lawler fought former WEC Welterweight Champion and future longtime Strikeforce Welterweight Champion Nick Diaz in his second UFC fight at UFC 47. In the second round, Lawler suffered his first of just two knockout losses in his career. Lawler subsequently lost again at UFC 50 to the late Evan Tanner, who would go on to win the UFC Middleweight Championship in the next bout.

On August 29, 2020, Lawler faced Neil Magny, who replaced Geoff Neal, at UFC Fight Night 175. A unanimous decision defeated him. Lawler was scheduled to face Mike Perry at UFC 255 on November 21, 2020. Lawler, however, was forced to withdraw from the fight due to injury.

Lawler will face returning veteran Nick Diaz in a rematch at UFC 266 on September 25, 2021. The fight was a five-round memorable non-title, non-main event battle at middleweight. Lawler knocked Diaz down in the third round after he refused to listen to the referee’s direction to go up to his feet. As a consequence, Lawler was declared the winner through TKO.

At UFC 276 on July 2, 2022, Lawler faced Bryan Barberena. He was knocked out in the second round of the back-and-forth fight. This bout won him the title of bout of the Night. Lawler was set to face Santiago Ponzinibbio at UFC 282 on December 10, 2022. However, Lawler was forced to withdraw from the tournament the week before owing to an unexplained injury.

Lawler faced Niko Price at UFC 290 on July 8, 2023. He knocked Price down 38 seconds into his farewell fight.
