Henderson began his wrestling career in high school, contending in 1987 and 1988 championships. In 1993, he competed in the NCAA championships. In 1991, 1993, and 1994, he was crowned University champion. In addition, he was the national senior champion in 1993, 1994, and 1997.

He represented the United States in the 1992 & 1996 Summer Olympics. In 1994 & 1997, he also participated in two World Wrestling Championships, finishing in 12th and 7th place, respectively. Throughout the 1990s, he coached wrestling at Cerritos College. Following his Olympic loss in 2000 and his trial for the World Team in 2001, he decided to pursue a career in MMA.

In 1997, Henderson began training in mixed martial arts (MMA), and he won his first competition at the Brazil Open. In his second UFC competition, he defeated champions including Allan Goes and Carlos Newton. Five of the 32 bouts in his third tournament, Rings: the King of Kings, were won by him.

In 2005 he claimed the welterweight championship after joining the Pride Fighting Championship late in 2000. On September 8, 2007, he competed for the heavyweight title upon his return to the UFC. He signed a 4th fight contract with Strikeforce in December 2009, but Jake Shields defeated him. He was successful in defeating other champions in subsequent contests. On October 8, 2016, he announced his retirement. However, on July 5, 2018, he competed in the UFC Hall of Fame and won.
