On February 7, 2009, Woodley made his professional debut against Steve Schnider. Headhunter Productions hosted it at the Holiday Inn Select Executive Center in Columbia, Missouri. Woodley won the opening round by submission. On April 30, 2009, Woodley faced Jeff Carstens in his second professional fight. In the first round, he won through submission.

Woodley made his professional debut against Jay Hieron on February 2, 2013, at UFC 156, replacing an injured Erick Silva. Woodley won the bout by KO in the first round after only 36 seconds. On June 15, 2013, at UFC 161, Woodley faced off against Jake Shields. Woodley was defeated in a split decision.

On November 16, 2013, Woodley fought Josh Koscheck in UFC 167. He won the bout through a first-round Knockout. He also received his first Knockout of the Night bonus award for victory. At UFC 171, Woodley challenged former Interim Welterweight Champion Carlos Condit. Woodley won by TKO after Condit injured his knee in the second round. Woodley’s new eight-bout deal with the UFC was revealed on April 8, 2014, alongside his fight against Rory MacDonald at UFC 174. A unanimous decision defeated him.
